Wilkes stu­died at the Roy­al Acad­e­my of Mu­sic, then played the or­gan at St. Da­vid’s Church, Mer­thyr Tyd­fil; Llan­daff Ca­thed­ral; and Monk­land church, near Le­o­min­ster (1860). He con­trib­ut­ed to the first edi­tion of Hymns An­cient and Mo­dern.
