Vandersloot was the son of physician Frederick William Vandersloot and Eliza E. Crouse, and husband of Cora Elwert. His grandfather was a German Reformed Church pastor, and a hymn writer.
His family moved to Clinton County, Pennsylvania, when he was a child.
In 1889, he began working at a music publisher in Williamsport, Pennsylvania. Within a decade, he had founded the Vandersloot Music Company, which he ran until selling the company in 1930.
For 28 years, he led the choir at the Pine Street Methodist Episcopal Church in Williamsport (James Black was one of the choir members).
