Penfield’s parents were Thornton Bigelow and Charlotte Devins Penfield, missionaries to India. After his father’s death in 1871, his mother moved the family to her parents’ home in Montclair, New Jersey.

Thornton attended St. Johnsbury Academy, Vermont, then returned to New York and lived with his mother and step-father while attending Columbia University in New York City. He then studied at Union Seminary, and worked for the New York Tribune.

After graduation from seminary, Penfield went to work for the Presbyterian Board of Home Missions, and later, the International Committee of the Young Men’s Christian Association (YMCA). He married Martha Lee Martin in 1894.
