William was the son of Alexander Hudnutt and Lydia Parker, and husband of Harriet Shew Beecher. His grandson, William Herbert Hudnut III, served in the U.S. House of Representatives, then for four terms as mayor of Indianapolis, Indiana (1976–92).
He graduated from Princeton University in 1886, and in 1962, became the oldest living alumnus of Princeton. Two of his sons and seven grandsons also attended Princeton.
In 1895, Hudnut was pastor of the Fish House Presbyterian Church, Northampton, New York. Both his sons and two grandsons were Presbyterian ministers like him.
