William was the son of William Perry Hamilton and Catherine Prince Roach, and husband of Zula Belle Doyle (married May 30, 1893, Sullivan County, Tennessee).
He lived in Lynchburg, Virginia, in 1910, and Roanoke, Virginia, in 1920.
He was educated at the Southern Baptist Theological Seminary, Louisville, Kentucky (ThM, 1893; ThD, 1904).
He served as the second president of the New Orleans Baptist Theological Seminary, then known as the Baptist Bible Institute (1928–42).
In 1920, Union University, Jackson, Tennessee, gave him an honorary Doctor of Laws (LL.D.) in recognition of his leadership and contributions to his denomination.
